Ο Λάκης Παπαστάθης γεννήθηκε στο Βόλο το 1943 αλλά μεγάλωσε στη Μυτιλήνη, όπου τελείωσε το Γυμνάσιο.
Σπούδασε σκηνοθεσία στο «Κέντρο Σπουδών Κινηματογράφου Αθηνών».
Γύρισε την πρώτη μικρού μήκους ταινία του (Περιπτώσεις του Όχι) το 1965 και τη δεύτερη (Ερμού 28) το 1968 (μαζί με τον Δημήτρη Αυγερινό), ενώ την περίοδο 1970-71 συνεργάστηκε - ως βοηθός σκηνοθέτης - με τον Αλέξη Δαμιανό στο γύρισμα της θρυλικής Ευδοκίας.
Η τρίτη μικρού μήκους ταινία του (Γράμματα από την Αμερική) κέρδισε το Α΄ Βραβείο στο Φεστιβάλ Ελληνικού Κινηματογράφου Θεσσαλονίκης, το 1972.
Από το 1982 ως σήμερα, γύρισε τέσσερις ταινίες μυθοπλασίας μεγάλου μήκους:

* Τον καιρό των Ελλήνων (1982) - Βραβεία Σκηνοθεσίας & Σεναρίου στο Φεστιβάλ Θεσσαλονίκης
* Θεόφιλος (1987) - Βραβείο Καλύτερης Ταινίας στο Φεστιβάλ Θεσσαλονίκης και επί-σημη ελληνική συμμετοχή στο διαγωνιστικό τμήμα του Φεστιβάλ Βερολίνου.
* Το μόνον της ζωής του ταξείδιον (2001) - Α΄ Κρατικό Βραβείο ταινίας μυθοπλασίας μεγάλου μήκους και άλλα επτά Κρατικά Βραβεία του Υπουργείου Πολιτισμού (στο πλαίσιο του Φεστιβάλ Θεσσαλονίκης). Επίσης, τρία Διεθνή Βραβεία και επίσημη συμμετοχή στο Φεστιβάλ του Τορόντο.
* Ταξίδι στη Μυτιλήνη (2010)

Παράλληλα με την κινηματογραφική του δραστηριότητα εργάστηκε επίσης - ως σκηνοθέτης και παραγωγός – στο χώρο της διαφήμισης και της τηλεόρασης.
Το 1970 ίδρυσε μαζί με τον Τάκη Χατζόπουλο την εταιρεία Cinetic που έγινε γνωστή για τις θαυμάσιες τηλεοπτικές παραγωγές της [Ιστορικό Αρχείο (1975-1976), Αναζητώντας την χαμένη εικόνα (1990-1997), Η αναβίωση του Αρχαίου Δράματος (1998)] και κυρίως για την πολιτιστική σειρά Παρασκήνιο (1976 – 2010) που είναι η μακροβιότερη εκπομπή της ελλη-νικής τηλεόρασης.
Συνεργάστηκε επίσης - σκηνοθετώντας τριάντα ταινίες μικρού μήκους που εντάχθηκαν σε παραστάσεις και συναυλίες - με το «Ελεύθερο Θέατρο», (Μια ζωή Γκόλφω -1974, Το τραμ το τελευταίο-1975) και με τον Διονύση Σαββόπουλο (Θίασος σκιών-1973, Χαίρω πολύ Σαββόπουλος-1976, Πρώτη του 2000 -1999).
